چکیده

Capillary electrophoresis (CE) techniques have been fundamental in advancing technology for the separation and identification of peptide and protein mixtures [1,2]. Today, they have become a staple in conjunction with mass spectrometry systems to resolve, identify and analyze protein and peptide characteristics with sensitivity and accuracy. Recent advances have miniaturized traditional CE instrumentation into microfluidic systems that can provide fast, highresolution separations with low sample volumes and better flow control [3]. However, the performance of traditional CE and microfluidic CE has rarely been compared. Furthermore, CE techniques such as on-chip gel electrophoresis, isotachophoresis [4], capillary isoelectric focusing [5] and capillary zone electrophoresis (CZE) have been applied from simple biomolecule separations to confirmatory diagnostics [6] and yet, rarely used for studying fundamental properties of peptides and proteins. In this way, CE has been underutilized as a stand-alone technique to probe structural and functional attributes of peptides.

Ionic strength effects on electrophoretic focusing and separations.

We present a numerical and experimental study of the effects of ionic strength on electrophoretic focusing and separations. We review the development of ionic strength models for electrophoretic mobility and chemical activity and highlight their differences in the context of electrophoretic separation and focusing simulations. We couple a fast numerical solver for electrophoretic transport with...

Improvement of Capillary Electrophoretic Enantioseparation of Fluoxetine by a Cationic Additive

One of the problems encountered in CE separations of basic compounds is the adsorption of analytes onto the negatively charged capillary wall which could lead to poor repeatability of migration time and peak area. Additionally, separation of enantiomers of chiral of basic drugs is commonly carried out in low pH buffer which contributes to strong ionic interaction of the cationic drug ions with ...

Electrophoretic mobility-ionic strength studies of proteins. IV. The effect of lipids on the electrophoretic patterns of human serum albumin at acid pH.

In previous publications from this laboratory, we have investigated the anomalous behavior of human serum albumin (l), various animal albumins (2) and human -y-globulin (3) in the Tiselius electrophoresis apparatus at acid pH. Because of increasing interest in the behavior of proteins in the acid region below their isoelectric point, this phenomenon has been receiving increasing attention from ...

Optimization of a nano-enzymatic reactor for on-line tryptic digestion of polypeptide conjugates by capillary electrophoresis.

This work aims at studying the optimization of an on-line capillary electrophoresis (CE)-based tryptic digestion methodology for the analysis of therapeutic polypeptides (PP). With this methodology, a mixture of surrogate peptide fragments and amino acid were produced on-line by trypsin cleavage (enzymatic digestion) and subsequently analyzed using the same capillary. The resulting automation o...
